Way of the Samurai 3 sẽ đưa bạn vào vai một samurai vô danh tại vùng đất mang tên Amana, đang được cai trị bởi vị lãnh chúa Fujimori Shuzen, vào thời chiến quốc loạn lạc.
Game sẽ có ba phe phái chính là nhà Fujimori, nhà Ouka – mang trong mình âm mưu lật đổ nhà Fujimori – và cuối cùng là những người dân tại làng Takatane. Người chơi sẽ được tự do lựa chọn con đường mà mình muốn và nó sẽ ảnh hưởng mạnh đến kết thúc của game (có hơn 20 kết thúc khác nhau).
Bên cạnh đó, bạn có thể rèn nên vũ khí cho riêng mình bằng nhiều mảnh nguyên liệu khác nhau và thông qua đó để học hàng trăm kĩ năng khác nhau tương ứng với từng loại vũ khí mà bạn cầm trên tay. Game cũng đem đến khả năng đánh giá thông qua các hành động của nhân vật chính, từ đó sẽ đề ra các danh hiệu phù hợp trong suốt cuộc hành trình.
