Darksiders II sẽ lấy bối cảnh ở cùng thời điểm với những sự kiện diễn ra tại phiên bản thứ nhất, khi War bị kết tội khởi đầu "Khải huyền" trước hạn định và trục xuất xuống Trái Đất bởi Charred Council. Khi hay tin và hiểu rõ War là một kỵ sỹ có danh dự cũng như lòng kiêu hãnh nhất trong cả bộ tứ, Death đã lập tức chống lệnh của Hội đồng và lên đường tìm sự thật, bằng mọi giá minh oan cho người anh em của mình. Để đi tìm câu trả lời và sự giúp đỡ, Death rong ruổi đến Nether Realms, nơi đứng giữa thiên đường và địa ngục, nơi được thống trị bởi những chủng tộc hùng mạnh nhất.
